How to Use a Fully Automatic Autoclave
1. First, remove the inner sterilization chamber. Then, add an appropriate amount of water to the outer chamber, ensuring the water level is level with the triangular shelf.
2. Replace the sterilization chamber and load the items to be sterilized. Do not overcrowd the chamber, as this may obstruct steam flow and affect the sterilization effect. 3. Ensure the mouths of the Erlenmeyer flasks and test tubes do not touch the walls of the sterilizer to prevent condensation from wetting the paper seal and seeping into the cotton plug.
4. Press down the lid and lock the valve. Wait until the set temperature and time display appears, then press the start button for sterilization.
5. After pressing the start button, the sterilizer will automatically heat up and cool down. Sterilization is complete when the temperature and pressure inside the sterilizer reach a safe range (temperature below 80 degrees Celsius). This experiment used 1.05 kg/cm², 121.3°C, and 20 minutes for sterilization.
6. After the required sterilization time, remove the sterilized items.
7. Place the removed sterilized culture medium in a 37°C incubator for 24 hours. If no contaminating bacteria grow, it is ready for use.
